摘  要:本试验采用酶法结合超声波辅助提取葛根异黄酮,通过单因素实验、正交实验和响应面实验研究了料液比、乙醇体积分数、超声时间、酶用量、酶解温度、pH值、酶解时间对葛根异黄酮提取率的影响。结果表明:葛根异黄酮提取的最佳条件:料液比是1:20、乙醇体积分数是60%、超声时间是30 min、酶用量是1.0%、酶解温度是60 ℃、pH值是3、酶解时间是1.5 h。在此条件下葛根异黄酮的提取率达到4.7%。葛根异黄酮对超氧阴离子自由基的清除率达到 57.1%,可以阻止Fe3+还原为Fe2+,故葛根异黄酮具有一定的抗氧化性。38930

Extraction and antioxidant activity of isoflavone from pueraria lobata
Abstract:In this experiment, the isoflavone was extracted from puerarin by the method of enzyme and ultrasonic. The material-liquid ratio, ethanol volume fraction, ultrasonic time, enzyme dosage, enzymatic temperature, enzymatic hydrolysis time, pH value on the extraction rate of isoflavones from pueraria lobata were studied by the single factor experiment, orthogonal experiment and response surface experiment The results showed that the optimum conditions for the extraction of total isoflavones of pueraria was as follows:liquid ratio was 1:20, enzyme dosage was 1.0%, the volume fraction of ethanol was 60%, the enzymatic temperature was 60 ℃, ultrasonic time was 30 min, pH value was 3, and enzymatic hydrolysis time was 1.5h. Under this conditions, the extraction rate of puerarin isoflavone reached 4.7%. The scavenging rate of the extraction on superoxide anion radical reached 52.7%, and extraction can prevent Fe3+ from deoxidizing Fe2+, which showed that the extraction of isoflavone has some antioxidant activity.

葛根是豆科植物葛的块根,含有葛根素、大豆苷、葛根木糖苷、大豆苷元等多种异黄酮类物质[1]。葛根的主要组成成分是葛根异黄酮,其总量约占葛根的5%~10%[2]。大量的临床实验证实葛根异黄酮具有广泛的药理功能,它不仅可以有效地防止高血压、有降血糖、而且还具促进血液循环、抗癌等作用[3-4]。葛根异黄酮还能增加女性雌激素的分泌,适用于女性的多个阶段[5-6]。同时,葛根异黄酮类化合物可作为活性氧的清除剂[7]。 葛根异黄酮的提取及其抗氧化性研究:http://www.lwfree.cn/shengwu/20190821/38016.html
